I have a Virginia Creeper which has now grown up the side of the house and is reaching the roof. Is this going to cause trouble on the roof tiles?

If it goes under the eaves and starts to creep into your roof space, then yes it could do damage. I would get a ladder and trim it back to a good 2 feet below the roof level. Trimming will do it no harm, we chop our neighbours encroaching nuisance every year and still it grows!
