I have made a wooden box 72"l x 8" wide x 9" deep and it will sit on stands against a wall. How many and diameter drain holes would suggest also what should I fill the box with ie compost/top soil or what % of each.

What are you planning to grow in it?

i would treat the wood and line the box with polythene firstly to keep the wood from rotting so quickly .

Good thinking NP. As you haven't said what you are wanting to grow it might be best to use ordinary potting compost intended for containers but remember that it only contains enough fertiliser to last six weeks.
